Description: This product is a specially formularized IL-1b Cell Stimulant solution. It is in an easy-to-use format. IL-1¦Á and IL-1¦Â are structurally related polypeptides that share approximately 21% amino acid (aa) identity in human. Both proteins are produced by a wide variety of cells in response to inflammatory agents, infections, or microbial endotoxins. While IL-1¦Á and IL-1¦Â are regulated independently, they bind to the same receptor and exert identical biological effects. IL-1 RI binds directly to IL-1¦Á or IL-1¦Â and then associates with IL-1 R accessory protein (IL-1 R3/IL-1 R AcP) to form a high-affinity receptor complex that is competent for signal transduction. IL-1 RII has high affinity for IL-1¦Â but functions as a decoy receptor and negative regulator of IL-1¦Â activity. IL-1ra functions as a competitive antagonist by preventing IL-1¦Á and IL-1¦Â from interacting with IL-1 RI. The human IL-1¦Â cDNA encodes a 269 aa precursor. A 116 aa propeptide is cleaved intracellularly by the cysteine protease IL-1¦Â-converting enzyme (Caspase-1/ICE) to generate the active cytokine.
Application Instruction:
1. Mix 200l of 50X IL-1b Cell Stimulant solution with 9800l of medium (for one 96-well plate). This diluted mixture must be used within the day. Note: Although the product is sterilized, user may need to filter the diluted solution with 0.2 filter to prevent any contamination during the dilution process.
2. Apply 100l of the above mixture to a well (96-well plate) and incubate cells at 37¡æ / 5%CO2 for 4hr~16hr. Collect the medium for further assays.
Storage Conditions: Store container at -20¡æ for six months or at -70¡æ for one year from date of receipt. Distilled 1X mixture must be used within one day.
